I just purchased ProSeries & have been using Turbo Tax Deluxe for many years. I just rec'd CD ProSeries Evaluation. It is very different. I was sent 2009. Do I transfer my 2009 TT to PS? Is there a manual to tell me how to do this? I don't know where

On the top tool bar towards the left click on file and then choose transfer. Under Option 1, use the drop down menu to choose transfer from 2009 Turbotax to 2009 ProSeries. Select the client files you wish to transfer or select transfer all.
Grafton provided you with good how to links for general operating information and Mimbreno gave you the how to for file transfers, now the best thing to do his hop on the software and give it a test drive. The best way to learn is to actually work with the software.
Create fictitious tax returns for practice. Use names that distinguish them from real returns, such Harry N Gouffy, Rollin A Long, Etta Lott and Olive Pitts.
Mimbreno gave you good instructions on the transfer process, and as others suggested set up a dummy client to learn the process. I have a client called "Dummy Doo Doo" whom is a close relative and it is good to learn the system and also complete "what if" returns

I believe that the Eval version of the program actually comes with test returns already in, but making new ones is even better, as you can learn to build it up right from the start
You might also want to replicate some 09 returns you've already prepared. It will help you become familiar with workflow and functionality. Try transfering some 08 Turbotax files to the 09 Proseries....
Proseries gives you the option of forms or worksheet entry.... I prefer forms entry but others will disagree... try both and see what you're most comfortable with....
Spend a lot of time with the help files.... there's a lot of good information and functionality.
The program is pretty robust so the learning curve is also..... try not to get to frustrated and remember the cardinal rule........................
